Is it just not working for THIS flash drive, or have you tried others? Have you tried your problem drive in another computer?
The drive could be bad (as in damaged from static shock) or it could simply have been corrupted by someone "unplugging" it from a computer without first going to the "safely remove hardware" icon to "stop" the drive before unplugging it.
If it's the second case, you can probably format the drive (it will wipe out all the data on it) but it will be usable afterwards. You will need to go into the disk manager to do this. Also, sometimes windows gets dumber than usual, and tries to assign the flash drive's drive letter to something that is already being used by a network drive, CD rom, etc. In that case, you can use the disk manager screen to change the drive letter of the flash drive to an un-used drive letter.
To get to the disk-manager right-click on your "My Computer" icon and go to Manage. You'll see the disk management section on the left side, about half-way down.

Is it shown in the Device Manager? If yes, just assign a new letter to it.
USB External Drive Does Not Show on Windows Explorer
If not, try a different computer. If it doesn't work on a different computer, it can be basically anything - contact oxidation, a loose contact, a broken PCB, a broken chip, a fried chip, whatever else...
