Doesn't sound like a software problem. Are you sure the USB port isn't working? Could it be the device that's plugged into the USB port which is malfunctioning?
I would troubleshoot as follows:
Go to Control Panel and into System settings and then to Hardware, and Device Manager. A window will pop up. Scroll to the bottom and hit the plus sign and you will see that the USB ports expand. If any have checkmarks or question marks, remove them by right clicking and choosing to remove them on the menu that pops up.
Once you remove them, reboot. It should re-install all the drivers for them. When done, reboot again and you should have working USB ports.
If this does not work, then reboot your computer. When it boots up again, immediately hit F2 or Delete, and it will pull up the BIOS settings. In there, scan around (do NOT CHANGE ANYTHING BUT THE PNP SETTINGS) to find the Plug and Play OS settings. Make 100% sure that's on a Plug and Play OS and that it's turned on.
If it does not work after that, then I suggest it's one of three other things:
1. Your USB port failed and it is an internal hardware issue.
2. Your USB supported item that you are plugging in is malfunctioning.
